Overview

IGT® UMI Adapter & UDI Primers is a series of kits used to detect samples with low-frequency mutations. Unique molecular identifiers (UMIs) are a type of molecular barcoding that tag each molecule in the library, which can reduce the false-positive rate and increase the sensitivity of mutation detection. iGeneTech offers 64 different 6-bp UMI adapters with edit distance not less than 3, which is enough to distinguish between original molecules and correct the errors during sequencing.


iGeneTech provides 384 combinations of different UDI primers for high-throughput sequencing in a single lane that reduces the index contaminations due to index hopping.


iGeneTech also offers Adapter & Adapter & UDI Primers without UMI technology.

High Accuracy and Sensitivity of Low-Frequency Mutation Detection

Test of low-frequency Horizon 0.1% cfDNA reference standard using IGT® UMI Adapter & UDI Primers successfully detected all 8 mutations with reliable mutation rate.

Significantly Reduce the False-Positive Rate

UMI can significantly reduce the false-positive rate by error correction. The following figure shows the raw data and corrected data with standard, SSCS and DSC de-duplication correction.


The DSC correction using UMI reduced the number of mutations with VAF less than 3% from 526 to 15, including all the 8 known mutations.
